Toyopearl AF-rProtein A-650F is a high capacity affinity resin designed for efficient capturing of monoclonal antibodies at production scale. The recombinant ligand is derived from one of the IgG binding domains of Protein A by optimizing amino acid sequence. It is expressed in E. coli, free of animal derived product. Multipoint attachment of this ligand to the rigid Toyopearl matrix reduces ligand leaching and enhances mechanical, thermal and alkali stability.

Typical static IgG binding capacity is > 45g/L and typical dynamic IgG binding capacity is > 30 g/L  (at 2 minutes residence time, 1 g/L loading concentration ). Cleaning-in-place (CIP) is possible with 0.1 M and 0.5 M NaOH. After more than 100 CIP cycles with 0.1 M NaOH at 16 min contact time typically more than 90 % of initial dynamic binding capacity is retained.

Chemical Stability of Toyopearl AF-rProtein A-650F IgG binding capacity is not significantly impaired by more than 100 CIP cycles with 0.1 M NaOH (red, 16 min contact time/cycle) and 0.5 M NaOH (grey, 17 min contact time/cycle). Dynamic binding capacity of hIgG determined at 10% breakthrough.
